b) Isocitrate dehydrogenase (IDH) mutation IDH is a rate-limiting enzyme in tricarboxylic acid (TCA) cycle which catalyzes Isocitric acid into [alpha]-ketoglutaric acid and reduces co-enzyme II nicotinamide adenine dinucleotide phosphate (NADPH) by oxidative decarboxylation.17 The IDH gene family has three isomerases, IDH1, IDH2 and IDH3.
For IDH activity assay, 1 mL reaction system containing 40 mM HEPES buffer (pH 8.2), 800 [micro]M NAD, and 200 [micro]M MnS[O.sub.4] was used, and the reaction was initialized with 2 mM isocitric acid. For cytosolic ACO activity assay, the enzyme extract was pre-incubated with an equal volume of 2 mM GSH for 1 h.
