Antibodies produce clumping of red blood cells or microbial cells because each basic antibody has two antigen-binding sites. Each antigen-binding site can attach to two antigenic determinants at once resulting in several antibody molecules binding with two cells.
For example, random chemical biotinylation can lead to either destruction of the antigen-binding site or the production of antibody fragments that bind to streptavidin in an orientation that is sterically unfavorable for antigen binding.
An alternative method is to screen peptide libraries to identify paratope-binding peptides, i.e., peptides binding to the antigen-binding site, and to compare the structures of these with that of the antigen (21,24-26).
