To account for this heterogeneity and facilitate accurate comparison with normal diploid cells, the number of exchanges visualised per metaphase spread was normalised for a diploid (2m) karyotype by multiplying observed SCE by 46 and then dividing by the observed chromosome number.
Elevated Endogenous SCE Levels in Primary Sarcoma Cell Lines.
All four commercially available sarcoma cell lines had endogenous SCE levels that were well above the expected normal range of 6-8, with median SCE counts of 15-31 per 2n metaphase spread (Table 1).
In the four primary sarcoma cell lines, where SCE analysis was repeated after approximately 4 months, the additional 5 to 15 culture passages did not appear to significantly affect the SCE levels.
Endogenous [gamma]H2AX Foci Are Increased in Sarcoma Cells but Do Not Correlate with SCE Levels.
